Chromocentres in polymorphs as interphase markers for chromosomes having increased constitutive heterochromatin.
نویسندگان
چکیده
After staining with the BSG technique for constitutive heterochromatin, human polymorphs exhibit chromocentres which are presumably derived from these chromosomal regions. In five individuals having an extended centromeric heterochromatin region on one No. 1 chromosome, a particularly large chromocentre was seen in the majority of polymorphs. Thus, prominent chromocentres in polymorphs may indicate the presence of constitutive heterochromatin variants.

Rumex acetosa Y chromosomes: constitutive or facultative heterochromatin?
Condensed Y chromosomes in Rumex acetosa L. root-tip nuclei were studied using 5-azaC treatment and immunohistochemical detection of methylated histones. Although Y chromosomes were decondensed within root meristem in vivo, they became condensed and heteropycnotic in roots cultured in vitro. 5-azacytidine (5-azaC) treatment of cultured roots caused transitional dispersion of their Y chromosome ...
متن کاملStatistical analysis of the three-dimensional structure of centromeric heterochromatin in interphase nuclei.
Translocation of genes into the pericentromeric heterochromatin occurs during cellular differentiation and leads to a long-term silencing of these genes. Consequently, a structural remodelling of this heterochromatin compartment is observed during differentiation but this remains to be defined from a topological point of view. In a previous study, we analysed the three-dimensional (3D) distribu...
متن کاملThe LINC complex contributes to heterochromatin organisation and transcriptional gene silencing in plants.
The linker of nucleoskeleton and cytoskeleton (LINC) complex is an evolutionarily well-conserved protein bridge connecting the cytoplasmic and nuclear compartments across the nuclear membrane. While recent data support its function in nuclear morphology and meiosis, its involvement in chromatin organisation has not been studied in plants. Here, 3D imaging methods have been used to investigate n...
متن کاملAlterations of constitutive pericentromeric heterochromatin in lymphocytes of cancer patients and lymphocytes exposed to 5-azacytidine is associated with DNA-hypomethylation.
BACKGROUND DNA hypomethylation plays a key role in carcinogenesis. The malignant transformation of cells as well as tumor progression is accompanied with increasing DNA hypomethylation in cancer cells. Nevertheless, the evolution of dis-epigenetic genomic alteration in the somatic cellular malignant transformation has not yet been clear. AIM To study the relationship between the pattern of ge...
متن کاملRNA-dependent stabilization of SUV39H1 at constitutive heterochromatin
Heterochromatin formed by the SUV39 histone methyltransferases represses transcription from repetitive DNA sequences and ensures genomic stability. How SUV39 enzymes localize to their target genomic loci remains unclear. Here, we demonstrate that chromatin-associated RNA contributes to the stable association of SUV39H1 with constitutive heterochromatin in human cells. We find that RNA associate...
